Edit: Ah, a fellow reader from r/ReLIFE has explained this nicely and I remembered about it now thanks to his explanation.
"It was referring to when the original plan for trying to get Ryo and Hishiro into a romantic relationship, supposedly she was going to be his future spouse but then since her experiment didn't go well the plan failed, future spouse here was referring the that plan."

tbh if we saw hishiron and ryo meet at the start i might have shipped them 
don’t get me wrong, i am totally on team kaizaki x hishiron and ryo x an BUT the way she woke up ryo had my heart going doki doki...the fact that they wanted to make ryo and hishiron a couple is still so fucking weird though so i’m glad that didn’t end up happening. also i noticed how we never saw hishiron’s face as an adult,
