Also not Yotsuba. There's no way she could completely change her outfit and makeup, and come running by with somebody else within seconds of 'Rena/Zero' disappearing.

Though I guess there's the possibility that 'Zero' isn't any specific one of them, but some kind of generic persona that all 5 sisters can assume. Or even less likely, but it's also possible that 'Zero' really is someone different but still somehow connected to the 5.

"Uesugi" is Fuutarou's and Raika's family name. There is no quintuplet with that name (yet).

To me she feels a lot like Ichika. I mean she has that adult like behaviour and wants to be an example for her sisters:

And then there's this strange bush movement in the last chapter. Perhaps from the conversation between Itsuki and Fuutarou, the girl in question (Ichika) remembered her meeting with him. There's even this reference to Ichika's question in the early chaps:https://mangadex.org/chapter/363853/5
Nino, Itsuki or Miku are highly improbable to be the ones, too much of hurdle on acting. My second choice is Yotsuba, she was my first choice in the beginning, but from her conversation with the other girl it didn't seem like she went missing while running.
There's only one thing for sure: Negi loves trolling us.

One thing is sure for me - the one that came to Uesugi dressed like that is one hell of a bitch. She came only to tease him but he's probably hurt because of that. If she wanna tell him her feelings she should've done it as herself or at least not lied while saying her name.

My guess is they aren't quintupletes but sextupletes instead just Rena or Zero was not much for the tightness the other girls have and so she and their parents decided that instead of six sisters it was five by her leaving the house with some other family of theirs. She meets fuutarou, talks about the five girls and then disappears. And now reappears asking about them so as to possibly take on their persona when she so desires to fool him in the future.
Or maybe it's Itsuki since she is the only other one apart from Miku and Nino that has long hair. Yeah I bet it's her.

Rei /零 = Zero

All of the quintuplets have a number on their name, by naming herself Rena she pretty much said she is the zero quintuplet, meaning thats a fake name.
