How about the hook to chin stuff? Can you really knock someone out that way?
 
Honestly the bigger issue with knockouts in fiction isn't where they hit (there are pressure points that work) but the length of time the people stay unconscious. The martial artists I've known who do that stuff started rousing their partner immediately and would get pretty damn worried if they weren't responsive after 20-30 seconds. Minutes or hours like you see in movies from that kind of knockout carries a pretty good chance of brain damage.
 
Aiming any kind of attack to the neck has a big change of injury. Unlike the head, it's unprotected by a strong thick skull protecting its inside. There's a good reason why hanging yourself is one of the most easiest way to end your life without suffering (assuming you die out of neck dislocation, not suffocation). If Video games' critical spot is the head, real life's critical spot is the neck.
 
@Dirtbug Looking at boxing, MMA/combat sports and other sport accidents (e.g. football, rugby injuries), it certainly is possible, but it has to be strong and accurate enough. I don't think I understand the mechanism, but it's probably straight-up concussion/blunt force trauma.
 
@Harambe Head is a critical spot. Skull is quite thin bone. Sure, it might seem strong if you just hit it with a fist (the bones in a hand aren't massive either), but if you have any kind of hard tool of more weight than paper mache, the skull is nothing much, not to mention a high energy penetrator like a bullet or shrapnel. Furthermore, even if the skull isn't broken, even a minor concussion will drop the target's capability immediately. Of course the neck is a serious spot as well if you can hit it.
 
as mentioned, if you've knocked someone out with a blow for more than like, 10 minutes, you might as well kill them, because they're not coming back in one piece
